Methods: Retrospective review of 87 patients with Glut 1 DS (45% female, age range 3 months35 years, average diagnosis 6.5 years) at Columbia University, from 1989 to 2010. Key Findings: Seventy-eight (90%) of 87 patients had epilepsy, with average onset at 8 months. Seizures were mixed in 68% (53/78): TGF-beta signaling generalized tonicclonic (53%), absence (49%), complex partial (37%), myoclonic (27%), drop (26%), tonic (12%), simple partial (3%), and spasms (3%). METHODS: This is a retrospective analysis from a single regional referral center on all patients undergoing TAB March 2003 through November 2010. Demographic, clinical, and surgical informations were recorded including changes in treatment based on biopsy results. RESULTS: In all, 237 Navitoclax clinical trial patients had complete documentation for review; the average age was 71 years (range 34 to 94) and 56% were women. Thirty-six patients had 42 positive biopsies; 26 biopsies were bilateral. PAR1 cleavage by FXa as analyzed with conformation-sensitive antibodies and a tagged PAR1 reporter construct was strongly enhanced if EPCR was available. Anti-EPCR failed to affect the tissue factor-dependent activation of FX, but high concentrations of FXa decreased EPCR-dependent protein C activation. Most importantly, the FXa-mediated induction of Erk1/2 activation, expression of the transcript for connective tissue growth factor and barrier protection in endothelial cells required binding to EPCR. Conclusions: Our results demonstrate that EPCR plays an unexpected role in supporting cell surface recruitment, PAR1 activation, and signaling by FXa.

Treatment with IL-17A/IL-17F or IL-22 expanded phosphoantigen 4-hydroxy-3-methyl-but-enyl pyrophosphate (HMBPP)-stimulated V gamma 2V delta 2 T cells from BCG-vaccinated macaques but not from naive animals, and IL-23 induced greater expansion than the other Th17-related cytokines. Consistently, Mtb infection of macaques also enhanced the ability of IL-17/IL-22 or IL-23 to expand HMBPP-stimulated V gamma 2V delta 2 T cells. When evaluating IL-23 signaling as a prototype, we found that HMBPP/IL-23-expanded V gamma 2V delta 2 T cells from macaques infected with Mtb or vaccinated with BCG or Listeria-actA prfA*-ESAT6/Ag85B produced IL-17, IL-22, IL-2, and IFN-gamma. Interestingly, HMBPP/IL-23-induced production of IFN-gamma in turn facilitated IL-23-induced expansion of HMBPP-activated V gamma 2V delta 2 T cells.
